About the Platform 

Well-known Italian Proteomics/Peptidomics Platform having International/National reputation and high- throughput/impact production.

Personnel - 1 Research Head; 5 Researchers; 2 Technicians; Various Postdocs and students.

Equipment - ESI-Q-Orbitrap-MS Q Exactive Plus, ESI-LIT- Orbitrap-MS Orbitrap XL ETD and ESI-LIT-MS LTQ XL ETD (all from ThermoFisher); MALDI-TOF-TOF-MS UltrafleXtreme (Bruker Daltonics); Proteineer fc II spotter (Bruker Daltonics); Easy-nanoLC (Proxeon); 2 NanoLC Ultimate3000 (ThermoFisher). Multipurpose equipment: several HPLC 1100 (Agilent); HPLC 1260 Infinity (Agilent); Nanodrop 2000 (ThermoScientific); different centrifuge equipment (Eppendorf and Thermo); UV-VIS-fluorescence multi-well reader.


Quantitative proteomics. Experiments on target cells after treatment with biotechnological products as well as on biotechnologically relevant organisms experiencing different pruduction conditions. Experiments up to 8-plex samples will be performed through digestion of whole proteomes, isotopically- labelled chemical tagging (based on Tandem Mass Tag – TMT reagents), peptide mixture rough fractionation, nanoliquid chromatography (nanoLC)-ESI-Quadrupole (Q)-Orbitrap-MS2 or nanoLC-ESI- Linear Ion Trap (LIT)-Orbitrap-MSn analysis, and dedicated bioinformatics.

Characterization of post-translational modifications (PTMs) in recombinant proteins. Recombinant products will be eventually purified by HPLC/SDS-PAGE, digested with a proteolytic enzyme and resulting peptide mixtures (eventually enriched for specific modified peptides) will be characterized by nanoLC- ESI-Q-Orbitrap-MS2, nanoLC-ESI-LIT-MSn or MALDI-TOF-TOF-MS.

Interactomics. Protein mixtures recovered from protein bait samples previously optimized for recovery and selectivity by the Consumer will be performed through proteomic shotgun experiments to identify specific protein interactors.

ISPAAM-Naples (Institute for the Animal Production System in the Mediterranean Environment) works on quantitative proteomics of organisms having biotechnological interest or of target cells after treatment with biotechnological products, characterisation of recombinant proteins with special focus on post-translational modifications (PTMs), interactomics of recombinant proteins from prokaryotic/eukaryotic sources.
